Strict whitelist with rejection message
From not in address1@some.net,address2@another.net,address3@yet.another.com
Reply with ^f, This mailbox only accepts messages from people in my address book. This message is an automated response regarding ^s, which was sent to ^R by ^f on ^T. If you did not send the above referenced message (^I), and if you do not want to add yourself to my addressbook, you've been framed by a spammer and you can trash this message.

I use store in Trash before Discard so I have a chance to look at them periodically.
The line with the addresses in it is limited to 500 characters, I think.
Strict blacklist
You could add a rejection message to this one like above.

You can sign for things with an alias to your email account, then when you're done with them you simply add the alias to this list and all future email to it will be trashed:

A lot of the newsletters I get have something in the subject line of every message to identify themselves. This is great for me, because I can let them through and automatically file them with a rule like this:
